Abstract
Statement of Purpose: Nature is the precious source of inspiration for the design of new versatile materials. Diverse structures in nature have concentric multilayers surrounding liquid core. The representative examples, such as hen’s eggs and mermaid purses which indicate protective egg case of oviparous sharks, indirectly verified liquid filled capsules with rigid membrane as the upmost strategy to preserve their tribe in the evolutionary aspect. Here, we developed multilayered polymer capsules with liquid core to create the ultimate boundary protecting encapsulated cells, which can flexibly respond to the external stimuli. The coacervation induced multilayered membrane can exhibit remarkable pH-responsive properties, forming tight structure against acidic pH and generating rapid degradation in basic pH, apt to be applied as oral delivery tools. These liquid core capsules were tailored as protective delivery vehicles for viral vectors and suspending cells, especially adeno-associated virus (AAV) and natural killer cells, to be used as the non-invasive treatment for intractable diseases.
