Single-cell RNA-sequencing of migratory cancer cells sorted by microfluidics: Discovering drivers of cancer metastasis

Abstract

Metastasis is the major cause of death in breast cancer patients, yet it remains challenging to pinpoint the key drivers in metastatic processes. We present a microfluidic cell migration platform enabling: (1) high-throughput migration experiments, (2) automatic image collection and analysis, and (3) reliable cell retrieval to enrich metastatic cancer cells. Using this platform, we have retrieved highly-migratory cells for in-vitro/in-vivo functional validation and single-cell whole transcriptome sequencing (RNA-Seq). In this manner, we identified new tumorigenic/metastatic biomarkers which were potential therapeutic targets.
